Fis蛋白質抑制插入序列IS2轉位之研究與大腸桿菌中調控蛋白GadX與微小核醣核?酸gadY抑制維生素B12受器btuB基因之轉錄

(I)本論文研究Fis(factor for inversion stimulation)蛋白質對插入序列IS2轉位機制的影響。經由Fis蛋白與IS2左右兩端DNA結合的電泳阻滯實驗、DNase I足印實驗(DNase I footprinting)、luciferase報告基因分析(reporter assay)與IS2轉位分析(transposition) 等實驗來證明Fis蛋白質會抑制插入序列IS2的轉位的頻率。 在電泳阻滯實驗上發現Fis蛋白質可與IS2左右兩端重複序列結合， 從DNase I足印實驗上發現Fis對IS2左端的結合區域是位於第44到第60核?酸，包含了IS2啟動子(p

romoter)的-35與-10之區間。而右端的結合區域是位於重複序列(inverted repeat)內側端點，與先前所發表之IS2轉位酵素(transposase) InsAB’的結合區相同。進一步分析Fis蛋白質與IS2啟動子結合與轉錄活性的關係，以IS2啟動子來表現luciferase報告基因(reporter) ，並置於有無fis基因表現的菌株來比較fis對IS2轉錄效率的影響。結果顯示Fis蛋白質可以減少IS2啟動子的活性達6倍。再分析IS2的轉位實驗， 我們發現在fis-突變株的大腸桿菌內其IS2轉位的效率比正常株減少了約64倍。從實驗結果發現Fis蛋白質可以結合在IS2左右兩端

序列，藉結合IS2啟動子而抑制IS2基因轉錄減低轉錄酵素產生，並干擾轉位酵素與右端重複序列結合區而抑制轉位的效率。(II)維生素B12的受器BtuB位於細菌的外膜上，除了運送維生素B12外它也是大腸桿菌素進入細菌體內的受器。一般對btuB基因的調控了解都是著重在於訊息RNA(mRNA)上二級結構的改變使得轉譯過程受抑制或促進，到目前為止尚未找到有調控蛋白直接參與在它的基因調控。為了找尋直接調控btuB基因的調控因子，我們從大腸桿菌基因庫中篩選對大腸桿菌素ColE7有較高耐受性的菌株。結果發現有一菌落對大腸桿菌素ColE7有較高的耐受性。經西方墨點法的檢測發現此抗性增強的菌株其BtuB蛋白質量減

少近90%。分離純化此菌株所攜帶之質體作核酸定序分析，結果發現此質體上帶有完整酸性調控因子gadX與gadY基因。因此推測gadX與gadY基因可能會影響btuB表現在。經電泳阻滯實驗與報告基因實驗，結果發現純化的GadX蛋白可以與btuB啟動子結合，gadX與gadY基因同時存在下會使btuB啟動子調控的LacZ比表現量減少近50%。從DNase I足印實驗確認了GadX與btuB的結合區域。再以不同酸鹼值的培養液比培養分析BtuB表現量，結果也顯示在pH 5.5酸性環境底下確實會影響BtuB的產量。自此確定了gadX與gadY有參與在butB基因的轉錄調控。
