前言:非傷寒沙門氏菌為人畜共通病原菌,可經由受沙門氏菌汙染的禽類蛋品或其製品感染人,造成沙門氏菌症。其中,雞傷寒沙門氏菌、雛白痢沙門氏菌和腸炎沙門氏菌經常可由雞隻中分離出,且帶有不同毒性因子。腸炎沙門氏菌屬於廣泛性宿主病原菌,可感染的宿主有牛、羊、豬、禽類、齧齒類及人類…等,而雞傷寒沙門氏菌與雛白痢沙門氏菌為對禽類專一性宿主的病原菌,雞傷寒沙門氏菌與雛白痢沙門氏菌遺傳背景相近,但雛白痢沙門氏菌僅能對幼雛造成高致死率,而雞傷寒沙門氏菌能對不同年齡的雞隻造成系統性疾病。華氏囊 (bursa of Fabricius) 為鳥類增生與分化B淋巴球細胞和產生抗體的器官,在抗體活化補體後,可對抗胞外的沙

門氏菌,移除華氏囊的雞隻,清除沙門氏菌能力降低。周邊單核白血球中單核球可經由胞飲、吞噬、趨化及釋出細胞激素來調節發炎反應,而禽類的異嗜球與哺乳動物嗜中性球相似,為第一線抵禦入侵的微生物。在吞噬細胞吞噬病原菌後,經由NADPH氧化酶將氧轉換為超氧化物,在先天免疫反應中,對抗沙門氏菌扮演重要的角色。目的: 評估雞隻對抗雞傷寒沙門氏菌、雛白痢沙門氏菌及腸炎沙門氏菌之血漿殺菌的能力與PBMC和PMN產生超氧化物的差異。材料和方法:將45週齡 (M1)、48週齡公雞 (M2) 和母雞 (F2) (品系為Hendrix) 隨機分為控制組、攻毒雞傷寒沙門氏菌S. Gallinarum (SG)、雛白痢沙門氏

菌S. Pullorum (SP) 及腸炎沙門氏菌S. Enteritidis (SE) 組,每次攻毒前抽血並分離雞隻PBMC與PMN且收集血漿,再以105 cfu菌數進行皮下注射,每三週抽血並攻毒,持續三次。於攻毒期間,測量雞隻體重。血漿部分,比較有無去除補體抗體力價與之殺菌活性,各試驗之差異: 1) IgG抗體力價趨勢: ELISA方法偵測血漿內專一性Anti-SG、Anti-SP和Anti-SE之IgG抗體力價。2) 抑制沙門氏菌生長: 攻毒前6週齡母雞10%血漿與菌數濃度為1,000 cfu/ml之SE、SG、SP或E. coli共同培養0、40、80和160分鐘後,塗盤培養至隔天,計

算菌落數。3) 血漿最小抑菌濃度:將血漿稀釋40%、20%和10% 並與同體積104 cfu/ml之SE、SG及SP混合均勻後,培養至隔天,測其OD595吸光讀值並計算出抑菌比例。在免疫細胞PBMC與PMN於攻毒期間,與SE、SG及SP (MOI: 100) 共同培養1、2及4小時,以Nitro blue tetrazolium (NBT) 試驗測量細胞產生超氧化物的情形。結果:攻毒期間,各實驗組與控制組相比,體重皆無顯著差異,顯示在此菌數攻毒不影響雞隻體重。血漿部分:1) 專一性IgG抗體力價: 攻毒SG後公雞與母雞抗體力價顯著提升,母雞攻毒SP後,抗體力價亦顯著提升,顯示攻毒SG較能

促進雞隻產生專一性IgG抗體,而有無去除補體活性,其ELISA吸光讀值亦有差異,與補體干擾有關。2) 抑制沙門氏菌生長: SG和SP能持續生長於攻毒前有無去除補體血漿,而SE與E. coli皆無法在有補體的血漿中存活。3) 血漿最小抑菌濃度: 攻毒SG後,其血漿同時含有抗體及補體時,其抑菌能力隨著攻毒次數增加而提升;攻毒SE組,去除補體活性血漿之抗體力價,與抑菌能力相似,而攻毒SP組,抑菌結果變化較大。PBMC部分:攻毒前,SE刺激PBMC產生的superoxide較高,且攻毒後,PBMC產生superoxide對抗SG、SP、SE及E. coli能力提升。但對抗不同菌株的時間點有差異。

M1攻毒SG的PBMC對抗SP、SE及E. coli時superoxide提升,M2則無此現象,母雞則於攻毒SG後,對抗SP及E. coli時superoxide增加,攻毒SP後,對抗SG及E. coli產生較高的superoxide。PMN部分:攻毒前PMN對抗SE及E. coli時superoxide較高,且M1 PMN產生superoxide受SG抑制,攻毒SE後,雞隻PMN 增加superoxide對抗三者不同宿主範圍的菌株及E. coli。結論:一、攻毒SG可幫助公雞與母雞產生較多的IgG專一性抗體。二、對禽類為宿主專一性之SG和SP可躲避其補體殺菌機制,但SE則無。三、攻毒試驗促使

雞隻產生記憶性抗體,且攻毒SG雞隻,其抗體亦可幫助補體抑制不同宿主範圍沙門氏菌。四、攻毒SE能增加雞隻PBMC與PMN之superoxide產量。

日常生活中會接觸到許多塑膠製品,鄰苯二甲酸酯類是一種添加在塑膠製品中的物質,由塑膠製品中會釋出鄰苯二甲酸酯類會造成人體或環境上的危害,2011年台灣爆發在飲品中添加鄰苯二甲酸酯類,造成恐慌,因此對於鄰苯二甲酸酯類的限制與控管日趨嚴謹,有鑒於此需建立一套簡單、準確、精密且低偵測極限的方法。本研究以分子印模磁性粒子結合氣相層析質譜儀,檢測六種常見的鄰苯二甲酸酯類,並建立分析方法。 研究結果顯示成功合成三層結構磁性粒子Fe3O4@SiO2@MIP,並應用其磁鐵粒子萃取六種鄰苯二甲酸酯類,其偵測極限介於0.015~0.004 μg/L之間,濃縮倍率為200~355倍。真實樣品添加1 μg/L與

5 μg/L標準品溶液,其回收率在83%~105%之間。 以分子印模磁性粒子結合氣相層析質譜儀萃取六種鄰苯二甲酸酯類,是一個簡單、快速和減低溶劑用量的萃取方法,且有效的降低偵測極限、提高濃縮倍率且可偵測低濃度樣品。
